Instruction of CNW vacuum SPE for operation and installation

Notes: Read the instruction carefully before start to operate vacuum device.

Introduction

The vacuum device is used for SPE sample pretreatment, filtration, and eluting etc. There are three different positions, 12 positions (2.CG1012.0001), 16 positions (2.CG1416.0001), 24 positions (2.CG1824.0001) are offered base on operating continuously with sample extraction and filtration, ensures simple pretreatment process and reduced experimental time consuming. The whole system contains a transparent glass chember and a head cover, the extraction can be achieved by controlling the flows of sample through SPE columns with vacuum condition. Different specification of collection tubes can be accommodated in glass slot by adjusting the stents inside, which includes glass and plastic test tubes, automatic sampling bottles, volumetric bottles and conical bottles etc, the lotion is usually drawn into sample collection tubes by selecting PP or stainless steel and Teflon guide needles. 12 positions system dries the lotion by importing flash air or nitrogen gas to collection tube, which ensures next analytical step. The drying device has a connection side which is able to attach SPE solvent inside. 12 positions also offers a PP tank for recycling waste liquid, ensures cleanness of glass chamber.

Installation

Install Cover, gaskets, guide needles and stopcocks:
1. Install four black legs to white cover. (Picture 1)
2. Note that the gaskets in the white cover should be kept in the correct position.
3. Attach the PP or stainless steel guide needles with luer connection side which located under device cover. (Picture 2)
4. Attach the stopcocks with luer connection side which located above device cover. (Picture 3)
5. Rotate the piston to keep it closed.

Adjust sample collection rack shelves:
1. The entire rack shelves consist of three support posts and a base plate. (Picture 4)
2. 12 position consists 5 collection rack shelves (with holes to carry collection tubes), 16 positions and 24 position consists 3 collection rack shelves. (Picture 5)
3. Selecting suitable collection rack shelves for your collection tubes (fits its size), install the whole system accordingly (Picture 6), adjust the height of the top plate to ensure the guide needles of SPE device would just pass through and stick into collection tubes.
4. In order to stabilize the whole rack shelves, a “c” type clip must be inserted on the groove side of support posts. (Picture 6)
5. The bottom collection rack shelve has stabilizing hole on it, when insert the collection tubes, they can attach each other and prevent shaking.
6. The stopcocks on the system is on the top which connects SPE columns, this ensure flow rate of extraction (starting or stopping point). (Picture 7

The usage of wasted solvent container:
The 12 position has its waste container, it (Picture 8) is a plastic tank which can be inserted into glass spot, it can restore large amount of waste solvent during extraction process, also ensures the cleanness of the whole glass chamber.

Connect Vacuum pump and Opening device:
1. Install a PTFE 1.0µm 50mm filter or surge flask between SPE device and vacuum pump.
2. Link vacuum pump to filter or surge flask with vacuum connecting tube; then link filter or surge flask to SPE vacuum device.
3. Open vacuum pump, using blow-down valve that near the vacuum meter on SPE device to adjust the vacuum degree (Picture10); the maximum degree can’t exceed 20 inches of mercury (508 mmHg), if not may destroy the device. The best vacuum degree for most analyte used on SPE columns is 3~5 inches of mercury. To easy pick and place collecting pipe support, blow-down valve also can control the vacuum degree.
4. The right operation of vacuum device includes: using blow-down valve adjusting vacuum degree to get optimum flow rate, meanwhile, the separate flow control piston also can adjust flow rate of each SPE column.
5. Under vacuum situation, removing SPE column need ensure flow control piston is closed. Remove head cover with pressure may cause drips splash or spill.

Optional Needles for Vacuum Manifold
Disposable Teflon Needles are designed to fit through the manifold lid via the luer fitting. These needles deliver the eluent directly from the SPE extraction column or cartridge into the collection vessel in the vacuum chamber. These needles, when used in conjunction with Teflon columns and Teflon frits ensure zero extractable from the column, frits, and fluid path. This combination is especially useful for critical sample analysis, such as environmental samples.

Summary

Vacuum degree cannot exceed 20 inches of mercury:
Blow-down valve and vacuum meter installed in glass guide channel, are used to control and monitor vacuum degree; Solvent-resistant PP luer connector on head cover can be joint with any SPE columns with male luer. Optional accessory Teflon anti cross contamination ensure no pollution of the whole flow path from head cover to collecting pipe. Effluent tank special applied to 12 position device is used to collect effluent before elution.

Sample filtration:
As samples pretreatment, purification and elution, samples collection and before sample injection need filtration treatment. Before filter the final eluent, take down the SPE columns from head cover, add a 25mm syringe filter into piston, then insert the SPE column into syringe filter’s female luer joint. After putting sample collection tube rack into glass guide channel, the whole device can be filtration treated. Take care the syringe filter cannot be installed before elution sample, if not may cause bubble block and result in that eluent cannot be passed away.
