Determination of Phenolic Compounds in Soil by SPE-HPLC Method

Abstract:A method for the determination of six phenolic compounds in soil is established which is extracted by Soxhlet's extraction, SPE and determined by HPLC. Optimization is conducted for the four main factors such as water sample pH,velocity of loading water sample, eluting solvent type and volume of eluting solvent that would affect recovery rate by orthogonal test. The order of influence on Soxhlet's extraction is water sample pH>velocity of loading water sample>eluting solvent type>volume of eluting solvent. Optimal results are determined as water sample pH 3,velocity of loading water sample 5mL/min, acetonitrile as eluting solvent and volume of eluting 10.0 mL. The extract is determined by HPLC and quantified by external standard method. The limits of quantification for all six phenolic compounds are 0.01~0.05 mg/kg in about l0.00 g soil samples, with RSD<10%(n=7) and the recoeries between 85.39%~105.82%. The results show that this method is sensitive,simple and suitable for the determination of phenolic compounds in soil.
